要旨Notch4 has been reported to be associated with schizophrenia. Rats with reduced g-secretase activity show behavioral abnormalities such as increased sensitivity to apomorphine.
Conditional deletion of RBP-J specifically in neuronal cells drastically reduced dopamine release in the striatum and caused a subtle decrease in the number of dopaminergic neurons.
RBP-J signaling regulates dopamine responsiveness in the striatum, which night suggest the possible involvement of Notch/RBP-J signaling in schizophrenia. However, it remains to be elucidated how Notch4 interacts with RBP-J and increases the risk of schizophrenia.
To elucidate the functions of Notch4 in the pathogenesis of schizophrenia, we performed behavioral analysis of Notch4-deficient mice and examined their responses to dopaminergic stimulation. We would like to discuss the possible roles of Notch4 in the pathogenesis of schizophrenia.
